Preparation of Solution of Paracetamol.

Paracetamol is an antipyratic drug that is generally used in the treatment of the fever .

The Quality of the Paracetamol is determined by the a method called uv specrtrometry where the standard solution of the paracetamol drug is prepared and then is compared with the normal drug or the drug of intrest for paracetamol drug in that product. 

Procedure for standard solution of paracetamol.

     1. Dissolve an accurately weighed quantity of paracetamol (100 mg) in small amount of distilled water and make up to 100 ml with distilled water to make Stock solution (1000 µg/ml).

    2. Further dilute 1 ml of this stock solution to 100 ml to produce second stock solution with concentration of 10 µg/ml.

    3. Pipette out 2, 4, 6, 8 and 10 ml from second stock solution and dilute it to 10 ml to make standard solutions having concentration of 2, 4, 6, 8, 10 µg/ml.

    4. Measure the absorbance of the solutions at 247nm using double beam UV-visible spectrophotometer against blank i.e., water.

    5. Plot a linear graph of absorbance vs. concentration (µg/ml)

 

Procedure for the saturated solution of the Paracetamol are as follows : -  

 

  1. Thoroughly clean all the necessary glassware with acid and wash 2 times with fresh distilled water.

    2. Take little amount of distilled water in a flask.

    3. Add excess amount of paracetamol in flask containing distilled water and stir it.

    4. After saturation keep the flask in a rotating chamber at 37°C at 60 rpm overnight.

    5. Measure the absorbance of the sample by UV-visible spectrophotometer.

    6. Determine solubility of paracetamol by using standard calibration curve.
